Domperidone is a peripheral, specific blocker of dopamine receptors. It is administered orally, rectally, or intravenously. Domperidone is given in order to relieve nausea and vomiting; to increase the transit of food through the stomach (as a prokinetic agent through increase in gastrointestinal peristalsis); and to increase lactation (breast milk production) by release of prolactin. It is also used in the scientific study of the way dopamine (an important neurotransmitter) acts in the body.
Treatment of Gastroesophageal Reflux Disease (GERD): Domperidone is used to manage symptoms of GERD, particularly those related to motility dysfunction, by improving the contraction of the stomach muscles and reducing the regurgitation of acidic contents into the esophagus.
Alleviation of Nausea and Vomiting: It is prescribed for conditions causing nausea and vomiting, including post-operative nausea and vomiting, chemotherapy-induced nausea and vomiting, and nausea associated with gastrointestinal disorders or medications.
Enhancing Lactation: In some cases, domperidone is employed to increase milk production in breastfeeding mothers who are experiencing difficulty maintaining an adequate milk supply. Its ability to stimulate prolactin secretion, a hormone responsible for milk production, is exploited for this purpose.
Prokinetic Action: Domperidone primarily functions as a peripheral dopamine receptor antagonist, particularly blocking D2 receptors in the gastrointestinal tract. This action promotes motility by accelerating the contractions of the stomach and intestines, aiding in the efficient movement of food through the digestive system.
