Function and Application
Vortioxetine is a potent serotonin reuptake inhibitor, with a high affinity to the 5-HT transporter (SERT) in the human body (Ki=1.6nmol·L-1), but almost no affinity to the norepinephrine transporter (Ki=113nmol·L-1) and dopamine transporter (Ki=1000nmol·L-1). Vortioxetine is also a 5-HT1A receptor agonist, a 5-HT1B receptor partial agonist, a 5-HT1D, 5-HT3 and 5-HT7 receptor antagonist, and a 5-HT uptake inhibitor. Animal studies have shown that in rats, vortioxetine increases the levels of serotonin, dopamine, norepinephrine, acetylcholine and histamine outside the cells of the ventral hippocampus and prefrontal cortex, which are brain regions related to depressive episodes, by acting on the above receptors together, while regulating the function of γ-GABA (γ-aminobutyric acid) and glutamatergic neurons, thereby exerting an antidepressant effect.
