Function and Application
MT is a metal-binding protein rich in cysteine. Its sulfhydryl group can strongly chelate toxic metals and excrete them from the body, thereby achieving detoxification. MT is related to the metabolism of trace metals. In all mammalian tissues, MT-1 and MT-2 are expressed synergistically. MT-3 is a specific member of the brain in this family. It can bind zinc and copper and has important neurophysiological and neuromodulation functions. Studies on many aquatic organisms have shown that MT plays an important role in the regulation of basic metal elements, and has inhibitory and detoxifying effects on non-basic metal elements. MT can effectively reduce the toxicity of heavy metals to the body by combining with heavy metals, and it is currently the most ideal biological chelating antidote in clinical practice.
Environmental protection
Utilizing the characteristics of the combination of MT and metal, it is possible to breed or use genetic engineering to establish a high expression system of MT to control heavy metal pollution. According to reports, tobacco transfected with yeast MT gene can significantly increase the absorption of cu2 in contaminated soil, and this method can be used to remedy areas contaminated by heavy metals.
MT and radiation resistance
Ionizing radiation can produce a large amount of free radicals, which can damage organisms directly or indirectly. Studies have shown that oral administration of MT can prolong the survival time of mice exposed to a one-time high-dose ionizing radiation, and reduce the damage to the immune system caused by one-time high-dose and multiple low-dose ionizing radiation. Oral MT can absorb a large amount of Cys that enters the body. It provides raw materials for repairing the disulfide bonds broken by radiation in the body.
MT and Parkison (PD) disease
Many experiments have proved that MT has a protective effect on the nervous system. Experiments have shown that in transgenic mice, overexpression of MT-1 can change the symptoms of encephalitis and promote brain repair. It is a protective factor in nerve cells. Through the study of rat stabbing model and ischemia model, it is found that MT-3 is involved in the repair of central nervous system damage. PD disease is caused by the induction of free radicals by 6-hydroxydopamine, and certain inducers of MT isoforms in the brain, such as oxidative stress, cytokines and inflammatory processes, can prevent this neurotoxicity, which is free from MT clearance Base related.
